After graduating from high school, Plaza went on to study film at the Tisch School of the Arts at New York University. While studying at NYU, she suffered a stroke at only 20 years old. The stroke caused her to experience temporary paralysis and expressive aphasia, which left her unable to speak or write. Thankfully, because she was so young, she made a full recovery. During her time at university, Plaza served as an intern at NBC, which afforded her the opportunity to work on Saturday Night Live. In 2004, she began performing at the Upright Citizens Brigade, a well-known theatre where she trained in improv and sketch comedy. Plaza also had the chance to perform stand-up comedy at the Laugh Factory and The Improv in New York City. She graduated from New York University in 2006.

Her first major role came when she was cast as comedian Seth Rogen’s love interest in the film Funny People in 2009.
